Mesenchymal Tumors

Mesenchymal Tumors arise from connective tissues, including bone, cartilage, fat, muscle, fibrous tissue, and blood vessels. This category covers mesenchymal tumors dincluding sarcoma, soft tissue sarcoma, and more. Sarcoma cell lines represent the malignant counterparts of mesenchymal tissues and are used for studying sarcomagenesis, metastasis, and drug resistance. Osteosarcoma cell lines (e.g., U2OS, Saos-2, MG63, HOS, 143B, SJSA-1, KHOS) are the most common bone sarcomas; they are used for p53 studies, chemotherapy screening (cisplatin, doxorubicin, methotrexate), and bone metastasis research. Chondrosarcoma cell lines (e.g., SW1353, CH-287, JJ012, OUMS-27) model cartilage tumors; they are used for studies on IDH mutations and collagen type II expression. Liposarcoma cell lines (e.g., SW872, 93T449, LS141, T449, LPS141) are adipocytic tumors; they are used for studies on MDM2 amplification and PPARγ differentiation. Rhabdomyosarcoma cell lines (e.g., RD, RH30, A204, Rh41, Rh18, Rh36) are skeletal muscle sarcomas; they are used for studies on PAX3/PAX7-FOXO1 fusions, myogenic differentiation, and chemotherapy. Leiomyosarcoma cell lines (e.g., SK-LMS-1, SK-UT-1, MES-SA, LMS-04) derive from smooth muscle; they are used for studies on uterine sarcoma and gastrointestinal stromal tumor (GIST) differential diagnosis. Fibrosarcoma cell lines (e.g., HT1080, SW684, HT1080 is widely used for invasion assays and matrix metalloproteinase studies) are fibroblastic sarcomas. Angiosarcoma cell lines (e.g., ISO-HAS, ASM, SVP-1) are vascular sarcomas; they are used for studies on endothelial malignancy and chemotherapy (paclitaxel). Ewing’s sarcoma cell lines (e.g., TC-71, A673, SK-ES-1, SK-N-MC, EW-7, RD-ES) are small round cell sarcomas with EWSR1-ETS fusions (most commonly EWSR1-FLI1); they are used for studies on targeted therapy (TK216) and EWS-FLI1 biology. Synovial sarcoma cell lines (e.g., SW982, HS-SY-II, SYO-1, Fuji, Yamato) have SS18-SSX fusions (SS18-SSX1, SS18-SSX2); they are used for studies on TLE1 expression and targeted therapy. Malignant fibrous histiocytoma (MFH, now often called undifferentiated pleomorphic sarcoma, UPS) lines (e.g., U-2 OS? no, U-2 OS is osteosarcoma; specific MFH lines like GCT, Hs 919.T, SU-CC-1) are used for studies on pleomorphic morphology and chemotherapy. Neurofibrosarcoma (malignant peripheral nerve sheath tumor, MPNST) lines (e.g., ST88-14, S462, MPNST-1, NF90-8) are associated with neurofibromatosis type 1 (NF1); they are used for studies on RAS/MEK/ERK pathways. Myxosarcoma lines are rare; some include MXY-1 and MyxoSarc. Mesenchymoma is a mixed tumor containing two or more mesenchymal components; cell lines are very rare. Undifferentiated pleomorphic sarcoma (UPS) lines are similar to MFH; they are used for studies on sarcomas without specific differentiation. Spindle cell sarcoma lines (e.g., SCS-1, KCS-1) are used for studies on spindle morphology and vimentin expression. Stromal sarcoma (e.g., endometrial stromal sarcoma lines, ESS-1, ESS-2) are uterine sarcomas with JAZF1-SUZ12 fusions. Mesenchymal chondrosarcoma lines (e.g., H-EMC-SS, MCS-1) are rare sarcomas with HEY1-NCOA2 fusions. Extraskeletal myxoid chondrosarcoma lines (e.g., H-EMC-SS, EMC-1, CH-287) have EWSR1-NR4A3 fusions. Researchers use sarcoma cell lines for studying mesenchymal differentiation and drug resistance. Osteosarcoma lines like U2OS are used for p53 studies and for testing chemotherapy combinations. Ewing’s sarcoma lines model EWS-FLI1 biology and are used for targeted therapy screening. Rhabdomyosarcoma lines are used for myogenic differentiation studies and for testing differentiation therapy.
