Knockout Cell Lines

Knockout cell lines provide defined loss-of-function models for studying gene activity in a controlled cellular background. By removing or disrupting a specific gene, researchers can directly evaluate its contribution to signaling pathways, disease phenotypes, drug sensitivity, cell growth, immune response, metabolism, differentiation, and other biological processes. Ascent Research offers CRISPR knockout cell lines across a range of commonly used mammalian cell line backgrounds. Our CRISPR KO cell line products are mainly generated using CRISPR RNP-based editing, in which Cas9 protein and guide RNA are delivered directly into cells. This non-viral strategy supports efficient gene disruption while avoiding unnecessary vector-based integration. Edited regions are typically confirmed by Sanger sequencing to verify the presence of target-site mutations and support reliable downstream use.

This category includes both clonal knockout cell lines and polyclonal KO cell pools. Clonal knockout cell lines are suitable when researchers require a more uniform genetic background and stable performance across repeated experiments. Polyclonal knockout cells, also called KO cell pools, provide a more economical option for early-stage screening, assay setup, pathway exploration, or projects where a mixed edited population is acceptable. These cell pools can help reduce cost and shorten project timelines before moving into single-cell clone selection.
