Reporter Cell Lines

Reporter Cell Lines are engineered to express a measurable signal (e.g., fluorescence, luminescence) in response to specific cellular events. This category covers reporter cell lines including fluoresence protein reporter and luciferase reporter. Our portfolio includes reporter cell lines for monitoring promoter activity, transcription factor activation, signaling pathways, protein localization, and cell lineage.

A reporter cell line stably integrates a reporter construct into the genome, ensuring consistent expression over many passages. Luciferase reporter cell line emits light upon luciferin addition, enabling high-throughput screening with excellent sensitivity and dynamic range. GFP reporter (green fluorescent protein) allows live-cell imaging and flow cytometry without additional substrates. RFP reporter (red fluorescent protein) is useful for multiplexing with GFP or for dual-color imaging. Luciferase reporter is the most sensitive for transcriptional activity, with firefly luciferase and Renilla luciferase being the most common.

Reporter gene assay measures changes in luminescence or fluorescence in response to drugs, ligands, or genetic perturbations. Reporter cells can be used for drug discovery (e.g., screening for agonists/antagonists of a pathway), pathway analysis (e.g., NF-κB, AP-1, STAT, Wnt, Notch, Hedgehog), or lineage tracing (e.g., Cre reporter lines). Fluorescent reporter lines like GFP or mCherry are common for cell sorting (FACS) and for studying gene expression dynamics. Bioluminescent reporter (luciferase) avoids phototoxicity and autofluorescence, making it ideal for long-term live-cell imaging.

Stable reporter lines are generated by integration into the genome, avoiding transient variability seen with plasmid transfection. Inducible reporter systems (e.g., Tet-On, Cre-Lox, light-inducible) allow temporal control of reporter expression. Signal transduction reporter cells monitor kinase cascades, second messengers (e.g., calcium, cAMP), or GPCR activation (e.g., CREB-luciferase).

Researchers use luciferase reporter cell lines for high-throughput compound screening, measuring IC50 values for pathway inhibitors. A GFP reporter line under a neural promoter (e.g., synapsin-GFP) tracks differentiation of stem cells into neurons. Pathway reporter cells like NF-κB-luciferase are used for inflammation studies (e.g., LPS, TNF-α). Our reporter cell lines are clonally selected for low background and high fold induction (often >100-fold), with validation by dose-response curves. Whether you need transcription factor reporter for signaling studies, dual reporter for normalization, or inducible reporter for controlled expression, our Reporter Cell Lines category supports your assay development with robust, reproducible tools.
