Reporter Cell Lines are engineered to express a measurable signal (e.g., fluorescence, luminescence) in response to specific cellular events. Stable reporter lines are generated by integration into the genome, avoiding transient variability seen with plasmid transfection. Inducible reporter systems (e.g., Tet-On, Cre-Lox, light-inducible) allow temporal control of reporter expression. Signal transduction reporter cells monitor kinase cascades, second messengers (e.g., calcium, cAMP), or GPCR activation (e.g., CREB-luciferase).

Researchers use luciferase reporter cell lines for high-throughput compound screening, measuring IC50 values for pathway inhibitors. A GFP reporter line under a neural promoter (e.g., synapsin-GFP) tracks differentiation of stem cells into neurons. Pathway reporter cells like NF-κB-luciferase are used for inflammation studies (e.g., LPS, TNF-α).
